Most Parkinsons disease (PD) cases are sporadic; however, about 10% of PD cases are inherited and many of the genes that are causative for these inherited cases are known. There is evidence that many of these genes play an essential role in the maintenance of normal presynaptic function. Mutations and sometimes overexpression of certain PD-causing genes lead to synaptic transmission defects and such defects may potentially lead to neurodegeneration. In this book chapter we present an overview of essential synaptic processes and link them to the PD genes that are thought to play a role in these synaptic processes.
