The establishment of a preclinical package for antiviral nucleoside phosphonate prodrugs

At the Laboratory of Medicinal Chemistry, two, structurally unrelated, series of nucleoside phosphonates prodrugs have been discovered that are both endowed with potent activity against the hepatitis B virus (HBV). Both compound classes are patent protected. In this grant proposal, a preclinical package will be assembled for both series of compounds. This includes the establishment of an improved synthetic procedure for large scale synthesis, broad antiviral testing, resistance profiling, ADME and toxicity studies and in vivo activity in relevant preclinical animal models. The generation of these data will allow to generate a full preclinical profile of the compounds that will be presented to pharmaceutical companies in order to outlicense the patent applications.
