Use of individual retention modeling for gradient optimization in hydrophilic interaction chromatography: Separation of nucleobases and nucleosides Vrije Universiteit Brussel
In this study, the separation of twelve nucleobases and nucleosides was optimized via chromatogram simulation (i.e., prediction of individual retention times and estimation of the peak widths) with the use of an empirical (reversed-phase) non-linear model proposed by Neue and Kuss. Retention time prediction errors of less than 2% were observed for all compounds on different stationary phases. As a single HILIC column could not resolve all peaks, ...

Nucleosides with Transposed Base or 4'-Hydroxymethyl Moieties and Their Corresponding Oligonucleotides KU Leuven Universiteit Gent
This review focuses on 4'-hydroxymethyl- or nucleobase-transposed nucleosides, nucleotides, and nucleoside phosphonates, their stereoisomers, and their close analogues. The biological activities of all known 4'-hydroxymethyl- or nucleobase-transposed nucleosides, nucleotides, and nucleoside phosphonates as potential antiviral or anticancer agents are compiled. The routes that have been taken for the chemical synthesis of such nucleoside ...
Stability studies of phosphoramidate nucleosides KU Leuven
Phosphoramidate nucleotides (as conjugates between nucleoside monophosphate and amino acids) have been developed as potential antiviral/antitumoral prodrugs and, recently, have been evaluated as substrate for polymerases. Despite of these intensive chemical and biological investigations, carried out with phosphoramidate nucleotides, little is known about their chemical stability and the influence of the nucleobase on the kinetics of the ...
Efficient and Accurate Potential Energy Surfaces of Puckering in Sugar-Modified Nucleosides KU Leuven
Puckering of the sugar unit in nucleosides and nucleotides is an important structural aspect that directly influences the helical structure of nucleic acids. The preference for specific puckering modes in nucleic acids can be analyzed via in silico conformational analysis, but the large amount of conformations and the accuracy of the analysis leads to an extensive amount of computational time. In this paper, we show that the combination of ...
Synthesis of 3 '-o-phosphonoethyl nucleosides with an adenine and a thymine base moiety KU Leuven
The synthesis and antiviral evaluation of new 3'-O-phosphonoethyl modified phosphonate nucleosides related to PMDTA and PMDTT is described. The reaction scheme starts from protected L-threose and the phosphonate group is introduced by the Arbuzov reaction. The Z-OH as well as the Z-deoxygenated nucleosides have been obtained. Unfortunately, none of these synthesized compounds shows activity against HIV and HCV
