Familial cortical dysplasia caused by mutation in the mammalian target of rapamycin regulator NPRL3 Universiteit Antwerpen
We describe first cousin sibling pairs with focal epilepsy, one of each pair having focal cortical dysplasia (FCD) IIa. Linkage analysis and whole-exome sequencing identified a heterozygous germline frameshift mutation in the gene encoding nitrogen permease regulator-like 3 (NPRL3). NPRL3 is a component of GATOR1, a negative regulator of the mTORC1 signaling pathway. Immunostaining of resected brain tissue demonstrated mTOR activation. Screening ...

Crystallization and preliminary X-ray diffraction analysis of kanamycin-binding β-lactamase in complex with its ligand Vrije Universiteit Brussel
TEM-1 beta-lactamase is a highly efficient enzyme that is involved in bacterial resistance against beta-lactam antibiotics such as penicillin. It is also a robust scaffold protein which can be engineered by molecular-evolution techniques to bind a variety of targets. One such beta-lactamase variant (BlaKr) has been constructed to bind kanamycin (kan) and other aminoglycoside antibiotics, which are neither substrates nor ligands of native ...
Multicentre laboratory validation of the colorimetric redox indicator (CRI) assay for the rapid detection of extensively drug-resistant (XDR) Mycobacterium tuberculosis Instituut voor Tropische Geneeskunde
Objectives To perform a multicentre study to evaluate the performance of the colorimetric redox indicator (CRI) assay and to establish the MICs and critical concentrations of rifampicin, isoniazid, ofloxacin, kanamycin and capreomycin. Methods The study was carried out in two phases. Phase I determined the MIC of each drug. Phase II established critical concentrations for the five drugs tested by the CRI assay compared with the conventional ...
Deafness induction in mice Vrije Universiteit Brussel
HYPOTHESIS: How to induce most efficiently severe sensorineural hearing loss in mice using a single coadministration of an aminoglycoside antibiotic and a loop diuretic?
BACKGROUND: The coadministration of aminoglycosides and a loop diuretic has been widely used to induce hair cell and spiral ganglion cell loss in guinea pigs. However, the development of new treatment strategies against sensorineural hearing loss, such as tissue ...
