Een van de kritische witte bloedcellen die een centrale rol speelt in het immuunsysteem zijn T lymfocyten. Ze worden gegenereerd uit bloedvormende stamcellen die een reeks afzonderlijke ontwikkelingsstadia ondergaan in een specifiek orgaan, de thymus genaamd. Dit orgaan verzorgt de correcte ontwikkeling van T cellen en dit resulteert in de vorming van twee types T cellen met een verschillende receptor op hun celoppervlak, ofwel een αβ ofwel een γδ T cel receptor. Beide cel types hebben belangrijke en unieke functies in het immuunsysteem en dit kan specifiek gebruikt worden in de context van immuuntherapie, zoals bijvoorbeeld voor de behandeling van kanker. Momenteel is er een gebrek aan kennis over hoe beide celtypes specifiek ontstaan vanuit eenzelfde voorlopercel. Dit willen we in dit project onderzoeken en dit is belangrijk om hun ontwikkeling selectief te kunnen sturen voor het gebruik in de context van celtherapie. Ons werk zal dus belangrijke nieuwe inzichten verwerven over hoe αβ en γδ T cellen ontstaan bij de mens wat belangrijk is voor de ontwikkeling van nieuwe cel therapie protocollen. To achieve this goal one needs to analyze molecules in their cellular environment and not under idealized test tube conditions. A major difference between in vivo and in vitro conditions is the crowdedness (and associated molecular complexity) of the cytoplasm and biological membranes. As a consequence diffusion in this environment is significantly slower. To make biology more quantitative, we should measure the key parameters such as reaction rates and diffusion coefficients in vivo. Here, I propose an experimental set-up to measure the diffusion of membrane proteins in live bacterial cells. The main objective of this project is to establish a relationship between the diffusion coefficient and number of trans-membrane helices (radius in the membrane) of membrane proteins. The mobility of fluorescently labeled proteins will be probed at the single molecule level in the membranes of a model prokaryotic organism – Escherichia coli. Single molecule tracking of membrane proteins systematically increasing in size (radius in the membrane) will be implemented to obtain a general model of protein diffusion in the membranes. Assigning actual numbers to the parameters of a cell will provide a quantitative context for systems biology efforts and sharpen our understanding of how a cell works. By training-through-research, the fellowship will allow me to learn new techniques (single molecule tracking, programming) and how to build/align microscope set-ups. This will help me transform from a microscope user to a microscope constructor and vastly increase my potential as a cell biologist. Their secretion is tightly regulated through Rab GTPase/effector complexes, including Rab27b-melanophilin (Mlph) and viral Gag-like proteins, including arrestin domain containing protein-1 (Arrdc1). We will use state-of-the-art density and size based AF4 fractionation technology to isolate monodisperse EV populations. Molecular signatures of each monodisperse population will be achieved through U+2013omics approaches. Preliminary evidence suggests the existence of small and larger EV, of which the larger EV are uniquely positive for Rab27b-Mlph and Arrdc1; in contrast small EV do not contain Rab27b-Mlph. Genetic interference with Mlph expression in hormone-sensitive breast cancer cells induces DNA double strand breaks and cell death. Recent reports indicate the importance of regulated EV release to control cytosolic double stranded dsDNA levels to avoid type I interferon induced cell death.
