Het tropische gewas vanille wordt geconfronteerd met een aantal bedreigingen die de toekomstige voorziening ervan in gevaar brengen. Vanille is een belangrijk gewas voor veel kleinschalige boeren in de tropen, en een zeer gewaardeerde specerij die wordt gebruikt in een uitgebreid scala van producten. Er is dringend behoefte aan het verbeteren van de weerbaarheid van de vanillesector tegen de verwachte effecten van klimaatverandering. Het huidige project is daarom gericht op het identificeren van klimaatbestendige vanillegenotypes en -teeltsystemen, door een interdisciplinaire aanpak toe te passen die distributiemodellering combineert met een experimentele evaluatie van soortspecifieke stressresistentie, en een participatieve beoordeling van de veerkracht van bestaande vanilleproductiesystemen. There is growing evidence, however, thatical and evolutionary dynamics can occur at the same time scale and can rongly interact with each other. Ignoring these interactions may amongst distort our view on human impact on natural ecosystems, for instance inntext of fisheries, land use, climate change, and the introduction of execies. In addition to the coupling of eco-evolutionary dynamics, we expelar interactions between social and evolutionary processes. Social behavd social structure may influence evolution, and evolution may shape socicture. Socio-evolutionary dynamics are expected in all organisms that shp behaviour, and will be especially important in species that form organcieties such as social insects and humans. Both eco-evolutionary and socutionary dynamics are therefore expected to be common and relevant to a nge of organisms and a broad array of fundamental and applied questions.p>The main objective of the research program is to obtain insight into tacteristics of eco- and socio-evolutionary dynamics and their impact on cal responses to environmental changes in a well-chosen set of model systrongly broadening the scale of earlier research. We will study eco-evoly dynamics in response to specific anthropogenic stressors (climate chanrient dynamics) in several key groups of aquatic organisms (zooplankton,c insects, fish, and their associated parasites) that belong to differenic levels, and thus firmly interact with each other through predator-preost-parasite interactions. We will also quantify eco-evolutionary interanbsp;in plant and microbial communities in response to changes in tempernd nutrient concentrations. For socio-evolutionary dynamics, we will notocus on social interactions within specific species (policing in sosects, quorum sensing in bacteria), but will also emphasize social inter that have impact on communities (communication in diverse microbianities, interspecific mutualistic interactions among plant, bacterial ant species). By studying eco- and socio-evolutionary dynamics on a broad f organisms within a consistent conceptual framework, we can explicitly the dynamics across groups to look for unifying principles. More specif we will identify key traits that impact the degree to which social, eco and evolutionary processes interact to impact ecosystem responses to nand anthropogenic stressors. Voor nieuw ontdekte activiteiten, inzicht in structuur, biosynthese en werkingsmechanisme van de bioactieve moleculen wordt beoogd met behulp van state-of-the-art benaderingen in de moleculaire microbiologie. Een tweede kleinere onderzoekslijn onderzocht de moleculaire mechanismen waardoor bodembacteriën pesticiden kunnen afbreken.Met behulp van genoomonderzoek willen we het enorme bacteriocinogeen potentieel van pseudomonaden verder ontrafelen door nieuwe toxine-immuniteitscombinaties te karakteriseren, waaronder hybride bacteriocines met gecombineerde toxinedomeinen. De bredere verspreiding over andere bodem- en plant-geassocieerde Proteobacteria (bijv. Van bijzonder belang is hun nieuwe werkingsmechanisme dat zich richt op de essentiële machinerie voor de assemblage van het buitenmembraan. Een derde type pseudomonad-bacteriocinen dat wordt onderzocht, zijn tailocinen, complexen met een hoog molecuulgewicht die evolutionaire verwantschap en structurele gelijkenis delen met verschillende bacteriofaagstaarten, indicatief voor meerdere onafhankelijke domesticatiegebeurtenissen. Van bijkomend belang zijn de extra genen die door sommige van deze elementen worden gerekruteerd, waaronder ook enkele reguliere en lectine-achtige bacteriocines.Naast een gediversifieerd bacteriocinogeen potentieel, vertonen pseudomonaden ook het vermogen om een breed spectrum van diverse secundaire metabolieten met antimicrobiële activiteiten te produceren. We onderzoeken de productie van dergelijke gespecialiseerde metabolieten door nuttige plant-geassocieerde Pseudomonas geïsoleerd uit de wortelomgeving van gematigde en tropische gewassen (maïs, tarwe, rijst, banaan). Er werd een bacteriocine-achtige specificiteit ontdekt voor het op salicylaat gebaseerde antibioticum promysaline, gesynthetiseerd door een rhizosfeerisolaat van rijst. They provide highly valuable ecosystem services that are threatened by sea level rise and changing sediment supply. Previous studies showed that the small-scale (order of m2) interactions between vegetation dynamics, water flow and sediment transport (so-called bio-geomorphic feedbacks) have a great impact on channel network formation and evolution at the landscape-scale (order of km2). We call this process bio-geomorphic self-organization. The aim of this project is to investigate, for the first time, the impact of plant species traits on biogeomorphic self-organization of intertidal landscapes. More specifically, we hypothesize that (1) different plant species traits lead to the self-organization of different channel network patterns, and (2) the resulting self-organized landscape structures determine the efficiency to distribute and trap sediments on the intertidal floodplain, and hence the resilience (adaptability) of the landscape to sea level rise and decreasing sediment supply.By using a combination of computer model simulations and field observations, we aim at producing new fundamental knowledge on landscape selforganization by bio-geomorphic feedbacks, and its implications for the resilience of intertidal landscapes against environmental changes." Ondanks tal van onderzoeken is men er nog steeds niet in geslaagd om te voorspellen of iemand zich goed of slecht aanpast aan verminderde beschikbaarheid van zuurstof (hypoxie). Dit project beantwoordt gedeeltelijk deze vraag door aan te tonen dat iemands genetisch profiel mede bijdraagt aan variabiliteit in hoogtetolerantie. Bovendien tonen wij voor de eerste keer aan dat enkele moleculaire adaptaties aan hypoxie deels onder invloed staan van genetisch factoren. Naast de niet-modificeerbare genetische factoren, kunnen externe omgevingsfactoren ook bijdragen aan de inter-individuele verschillen in hoogtegevoeligheid. Wij bestudeerden de invloed van eennutritionele factor, m.n. nitraatsupplementatie, en toonden aan dat deze de negatieve effecten van hypoxie op prestatiecapaciteit deels kan tegengaan. Inzicht in de fysiologie aan de grenzen van menselijke tolerantie voor hypoxie is een waardig doel op zich, maar kan bovendien naast eengezonde populatie, ook voor een patiëntenpopulatie van groot belang zijn. "Unravelling the regulatory roles of defective invertases in plants" "Wim Van den Ende" "Moleculaire Biotechnologie van Planten en Micro-organismen, Biochemie, Moleculaire en Structurele Biologie, Biokristallografie" "Mankind is becoming increasingly dependent on plant production, not only for staple foods, but also for energy, fine chemicals, functional foods, drugs and vaccines. Four KULeuven labs have joined forces to contribute to the development of superior crops with higher yields and biomass through optimization of carbon allocation and stress tolerance. Plants use glucose and sucrose #signals# to coordinate metabolism with environmental and hormonal signals for optimal growth and development. Although sucrose-specific signalling pathways exist in plants, the sucrose sensor(s) involved in the process have not yet been identified. Invertases bind and degrade sucrose and play an important role in carbohydrate partitioning. The discovery of several catalytically defective invertases suggests that they could act as sucrose #sensors# or fulfil other regulatory roles. This project aims at unravelling the exact role of defective invertases in plants. In particular, it will be investigated whethersucrose can bind to these invertases by generating sucrose-protein complexes, solving their three-dimensional structure using X-ray crystallography as well as by docking and molecular dynamics simulations. These studies will be complemented by protein interaction and cellular and whole plant molecular, biochemical and physiological studies of sucrose signalling in Arabidopsis thaliana knock-out mutants and plants overexpressingwild-type and mutant proteins."
