The VAGABOND ITN, consisting of 12 academic and 6 non-academic partners from 8 European countries, aims to create a multidisciplinary and multi-sectoral program to validate new therapeutic interventions in paediatric cancer. This network will be sustainable embedded in the ITCC (Innovative Treatments for Children with Cancer) Consortium. The VAGABOND ITN will focus on 15 cutting-edge research projects studying molecular, epigenetic and immunological targets. All projects will (partly) include target identification, in vitro and in vivo validation, compound testing and preparation for clinical implementation. This consortium will highly stimulate international education and multidisciplinary exchange of expertise in the different steps of pre-clinical drug development. This setting provides an opportunity for a unique European training network in which early stage researchers are equipped with a broad knowledge in pre-clinical drug development. Through this network students will gain access to other leading academic research groups, specialized multi-sectoral companies and the most innovative techniques and insights in the complexity of translational medicine to prepare them for future challenges in their career pathway. We have assembled a strong consortium with ten expert clinical centers, access to large volumes of the required tumor types. IMMUcan will mobilize the majority of academic trials running or expected to start recruiting patients during the project period. A centralized workflow for samples, via a state-of-the-art biobank will increase reproducibility as all tissues will be processed and stored in a uniform way, following proofed SOPs. The project will perform in depth immunoprofiling with cutting edge technologies including CyTOFF, single cell RNA-seq, peptidogenomics or microbiome analysis. IMMUcan will analyze the data in order to understand the host/tumour interaction in the absence of treatment (naïve population) and with treatment (patients in follow-up) to identify potential predictive biomarker for response to immunotherapy, or develop rationale for combination therapies. IMMUcan will provide an IT platform and legal/ethical contractual framework where participants from both academia and industry can pursue their own independent investigations utilizing the IMMUcan data and effectively testing and improving the functioning and relevance of the database.  A sustainability plan will be developed to ensure the collection of follow-up data and the accessibility of the data platform. In a series of breakthrough clinical studies, the hosting lab has shown that chemotherapy given during pregnancy is relatively safe for the infants in the short term. However, children born after in utero exposure to chemotherapy are often small for their gestation age, present with lower IQ scores and have higher risk to wear glasses, suggesting possible long-term health effects.Because chemotherapy elicits (epi)genetic alterations to cancer patients, we hypothesize that chemotherapy-mediated long-term health effects could be due to (epi)genetic changes in the offspring of women treated with chemotherapy during pregnancy. Thus, SCAR aims to determine whether in utero exposure to carboplatin may be linked to DNA damage in the offspring. My objectives are to 1) identify the genetic damage at single cell level and 2) determine epigenetic modifications; in umbilical cord blood cells of children in utero exposed to carboplatin. To this end, I will integrate highly innovative sequencing techniques and the access of the hosting team to a unique registry of pregnant cancer patients.As the phenotypic expression of these genetic alterations could take decades to appear, studying the underlying molecular epi(genetic) changes at birth, will provide more immediate evidence than long-term clinical follow-up, and will be instrumental to define safe and solid treatment guidelines for pregnant cancer patients. Om dit te overkomen, wordt uitgekeken naar de rol van nanomaterialen (NMs), zijnde kleine (nm schaal grootte synthetisch vervaardigde materialen) waar verschillende reeds gebruikt of verder getest worden voor klinische toepassingen. Het gebruik neigt vooral als dragers voor pharmaceutische agentia of als middel voor fysieke antikanker methodes (bvb lokale opwarming). Tot op heden is het succes van deze NM nog tamelijk beperkt, wegens de geringe specifieke afgifte van NM in tumoren, en het gebruik van de NMs louter als dager van agentia met intrinsieke problemen (bvb resistentie).Om deze problemen te overkomen, richt dit project zich op het gebruik van bepaalde types metaal-gebaseerde NMs die spontaan oplossen in een biologische omgeving (Fe-gedopeerde ZnO, CuO of silver NMs van verschillende groottes en opervlakte coatings). Door de snelheid van oplossen te controleren, kunnen NMs verkregen worden met een selectieve toxiciteit tegenover kankercellen als een nieuw and algemeen bruikbaar antikanker middel dat:1. een hogere therapeutische efficiëntie bezit tegen moeilijk te behandelen (resistente) cankers2.  een hogere specificiteit van de therapie door het intrisieke grotere effect van de NMs op kankercellen tegenover niet-kanker cellen.Om een betere afgifte van de NMs in de tumoren te verkrijgen, zullen cellen gebruikt worden die van nature uit spontaan naar de tumor migreren. The predominance of aggressive Type II tumours, which are characterised by a high frequency of p53 mutations, and primary or acquired resistance to platinum-based chemotherapy profoundly contribute to the high mortality rate. With current standard therapy the median overall survival of metastatic platinum-resistant (Pt-R) ovarian cancer patients is only 14 month. There is a pressing need for more effective, innovative treatment strategies to particularly improve survival in this subgroup of EOC patients. The GANNET53 trial aims to achieve this goal by applying a highly innovative concept that has grown from solid basic research findings made by members of the GANNET53 consortium. This is a drug strategy targeting a central driver of tumour aggressiveness and metastatic ability, namely mutant p53, via an innovative new Hsp90 (heat shock protein 90) inhibition mechanism. The most advanced, second-generation Hsp90 inhibitor will be used, Ganetespib. The first part (Phase I) of the GANNET53 trial will test the safety of Ganetespib in a new combination with standard chemotherapy (Paclitaxel weekly) in Pt-R Type II EOC patients. The second part (randomised Phase II) will examine the efficacy of Ganetespib in combination with standard chemotherapy versus standard chemotherapy alone in EOC patients with Pt-R Type II tumours. We established a highly efficient consortium with previously proven capability and manpower to perform this multicentre clinical trial and assess our innovative therapeutic concept in this deadly disease. Our consortium consists of national clinical trial groups in gynaecological oncology and high-volume University centres as well as noted p53 scientists and 3 innovative SMEs. Responding to this imperative in the context of both the broad Virtual Physiological Human (VPH) initiative and the paradigmatic cancer domain, CHIC proposes the development of a suite of tools, services and secure infrastructure that will support accessibility and reusability of VPH mathematical and computational hypermodels. These will include a hypermodelling infrastructure consisting primarily of a hypermodelling editor and a hypermodelling execution environment, an infrastructure for semantic metadata management, a hypermodel repository, a hypermodel-driven clinical data repository, a distributed metadata repository and an in silico trial repository for the storage of executed simulation scenarios. Multiscale models and data will be semantically annotated using the ontological and annotating tools to be developed. An image processing and visualization toolkit, and cloud and virtualization services will also be developed. The CHIC tools, services, infrastructure and repositories will provide the community with a collaborative interface for exchanging knowledge and sharing work in an effective and standardized way. A number of open source features and tools will enhance usability and accessibility. In order to ensure clinical relevance and foster clinical acceptance of hypermodelling in the future, the whole endeavour will be driven by the clinical partners of the consortium. Als in de hele EU een hoge vaccinatie couverture wordt behaald, kunnen we alle cervicale, anale, vaginale, penis-, hoofd- en nekkankers veroorzaakt door HPV zogoed als volledig elimineren. Momenteel zijn er nog ongeveer 67 500 nieuwe gevallen per jaar in de lidstaten. Een belangrijke uitdaging voor het bereiken van een hoge vaccinopname is het overwinnen van de ""vaccin-aarzeling"", vaak gebaseerd op onzekerheid over de veiligheid van het vaccin. In de meeste landen is de opname momenteel te laag om groepsbescherming te bereiken. Het PROTECT-EUROPE-project heeft tot doel dit probleem op twee niveaus aan te pakken. Ten eerste zal het informatie en training aanrijken over het optimaliseren van één-op-één communicatie met jongeren en hun ouders / verzorgers voor het brede scala aan gezondheidswerkers die betrokken zijn bij HPV-vaccinatie. De opleidingsprogramma's zullen online worden aangeboden en zullen via een ""train-the-trainers"" benadering in de lidstaten worden opgenomen. Ten tweede zal het project begeleiding beiden aan, de lidstaten en maatschappelijke organisaties een rol op het gebied van de volksgezondheid, aan de hand van een reeks campagne-instrumenten gericht op jongeren en hun ouders/verzorgers die kunnen worden gebruikt om vaccinatie aan te moedigen. De campagnetools zullen worden ontworpen voor gebruik op verschillende platforms, waaronder sociale media, websites, posters en folders. Het potentieel van sport, met name voetbal, als influencer zal worden verkend. Bijzondere aandacht zal worden besteed aan de aanpak van kwesties van gelijkheid en diversiteit.
