GENERATION OF ARTIFICIAL NUCLEIC ACID SYSTEMS FOR BIOTECHNOLOGY AND THERAPEUTICS

1、Design and preparation of artificial DNA and RNA molecules. Search for the optimal structure. Synthesis, purification and characterization of sequences. Optimization of PCR and reverse transcription reaction conditions. 2、Enzymatic methods screening for XNA assembly. Selection of the optimal enzymes (XNA-dependent nucleases, ligases, and polymerases) for cloning and assembly of genomic XNA fragments and vectors. Analysis of the product. 3、In vitro examination of XNA constructs. Search and verification of optimal in vitro models. 4、In vivo examination of XNA constructs. Search and verification of optimal in vivo model cell cultures. Optimization of XNA/rXNA template transformation and cell growth. Analysis of transformants. 5、Writing the scientific publications and Ph.D. dissertation Collection and processing of scientific results and published materials.
