Improving insight into colonic drug disposition for the development of predictive tools.
Colon targeted delivery of drugs after oral intake is the preferred approach for local treatment of high burden bowel diseases including ulcerative colitis and colorectal cancer. However, the success of colonic targeting is hampered by a poor understanding of the behavior of relevant drugs and formulations along the gastrointestinal tract and, consequently, an erratic prediction of their performance in humans. The present research project pursues a unique and holistic understanding of colonic drug disposition in humans by combining (i) the expertise of Drug Delivery and Disposition (KU Leuven) in absorption profiling, (ii) the skills of the Gastrointestinal Endoscopy unit (UZ Leuven) in assessing the human colon, and (iii) collaborations with experts from the University of Athens and Uppsala University. The obtained fundamental knowledge will serve as reference to evaluate the predictive power of current in vitro and in silico simulation tools and drive their further improvement.
